Histamine Analysis

Screening, Quantitative, and Semi-Quantitative

Whether its fresh off the boat, or further processed, RMD offers a number of solutions to Histamine testing, that are built on many years of experience in clinical diagnostics. Quality is the cornerstone of any business, and we are here to help you reinforce that commitment to quality with cutting-edge technologies.
  • HistaSure- Dipstick-  Rapid Dipstick test for the semi-quantitative detection of histamine in seafood
  • HistaSure- Fully Quantitative - Rapid ELISA for fully quantitative detection of histmine in food

HistaSure - Dipstick

The worlds first production dip-stick for the detection of Histamine in Fish is here! In Just minutes you can screen for the presence of histamine. We pre-set the cut to international standards of 50ppm, but the system is easily changed to allow for adjustable cut-offs in order to meet your QC needs.

Instructions:

  1. Mix 10 grams of tuna with 240 mL of water
  2. Add 100 μl of the sample to the tube containing 500 ul of Acylation Buffer - Vortex to mix
  3. Add 100 μl of the acylation reagent to the tube in step 2
  4. Vortex to mix and incubate for 5 minutes
  5. Pipette 100 ul of the acylated sample to the Sample Diluent.
  6. Vortex to mix Add 100 ul to the cassette
  7. Read results after 5 minutes.

Advantages

  • Fast results in minutes
  • No special equipment needed
  • Easy interpretation
  • Can be performed Dockside
  • Reuse the sample in our ELISA

Results and Interpretation:

The visual inspection of the LF-cassettes can give the following results: 2 lines or 1 line.

2 lines: The histamine concentration of the sample is below 50 ppm; the sample has passed. Example: 2 lines are visible - the sample has passed

2line-histasure

1 line: Only the control line is visible. The histamine concentration of the sample is above 50 ppm; the sample has failed and further investigations have to be performed. Example: 1 line is visible – the sample has failed.

HistaSure - Fully Quantative

New and improved ELISA method that offers fully quantitative results in 30 minutes! With a simple water extraction and our improved ELISA method, we have surpassed similar methods in speed and usability. Ideal for high volume testing, or small runs, this new ELISA is a breakthrough.

Instructions:

  • Mix 10 grams of tuna with 240 mL of water
  • Add 50 μl of the sample to the running buffer
  • Add 50 μl of the acylation reagent to the tube in step 2
  • Vortex to mix and incubate for 10 minutes
  • Pipette 50 µl from step 3 into each well, and add 100 µl of Antiserum conjugate, incubate for 10 min.
  • Decant and wash 3 times
  • Add 100 ul of Substrate and incubate for 10 minutes on a shaker
  • Add 100 µl of Stop Solution, and read the plate at 450 nm (reference 620 – 650 nm)

Advantages

  • Fast results in minutes
  • Wider range from 2.5 – 300 ppm
  • No centrifuge required
  • Easy interpretation
  • Less hands-on time.
  • No toxic organic solvents like methanol
  • No additional sets of tubes needed
